During the time we were searching for ideas, we found out that Crohn’s disease makes a lot of people’s lives difficult with its serious effects in the gastrointestinal tract. Thus we have made a resolution to engineer a genetically modified probiotic bacteria, which could reduce the disease’s symptoms by producing a substance called N-acetylglucosamine from natural chitin resources. These bacteria could be transplanted into the human intestines, by the means of yoghurts or capsules. With this method it would be much easier to treat patients with Crohn’s disease in the future.<br><br><br><br></span></div>

We decided to start two projects. Both of them is coconnected with nutrition and he gastrointestinal tract.

In the first project our main aim is tocreate a bacterium which produces chitinase enzyme to treat Crohn's disease, the other is an Amanitin-biosensor. With the latter, our aim is to detect the toxin of the poisonous death cap.

Crohn’s disease is a chronic, festering, inflammatory intestinal disease, which causes diarrhoea, ulcerous wounds in the large intestines. It is unknown how it develops in pour body and its cure is unknown as well. Researches have proved that the N-acetylglucosamine is able to ease the symptoms.

The polymer form of N-acetylglucosamine is chitin. The chitinase enzyme converts chitin to its monomers.We plan to insert the chitinase coding gene into bacteria. Thus taking the bacteria as a probiotic they can produce their effect locally and for longer time.

We have weekly sessions on tuesdays where we discuss what we have done so far, and what to do afterwards. Every member of the team gets new tasks to do until the next meeting. We have lessons in the mornings during the weekdays, so we can only work on our project in the afternoons and on weekends.

We have split into two groups. One is working on the Amanitin-biosensor, the other one is doing research on Crohn's disease. Our specialists on the disease are: Petra, Réka, Andris and Martin. The subteam making the biosensor consists of Miklós, Gergő, Márk and Ákos. The leader of the team is our teacher, Mr Sándor Bán.

The Crohn's disease is a chronical, festering, intestinal disease. The whole intesine is sappurated, it's incurable. Our aim with the second project is protection in simple and cheap way with N-acetylglucosamine. Animal cells doesn't have a periplast, only an extracellular matrix surrounds the cells in order to protect them. The N-acetylglucosamine connects to this matrix increasing its protective effect. As the N-acetylglucosamine's polymer form is the chitin, therefore we can produce chitinase enzyme with a bacterium which is also an intestinal one which can disassemble the chitin to N-acetylglucosamine.

Nowadays, death cap (Amanita phalloides) is one the most responsible species worldwide, which cause poisoning and significant rate of these toxications (mostly those which occur far from the necessary medical attendance) has lethal outcome. In Califonia, for example, 6300 poisonings were registered between 1993 and 1997. The main cause of these accidents is that the death cap may be confused with Agaricus species, due to their similar apperance, thus threatening verdant and negligent people. With our project it would become easier to distinguish Amanita from other species, so ensuring the mushroom we eat is not toxic.

Welcome! This is the first Hungarian iGEM HS team's homepage. During the time we were searching for ideas, we found out that Crohn’s disease makes a lot of people’s lives difficult with its serious effects in the gastrointestinal tract. Thus we have made a resolution to engineer a genetically modified probiotic bacteria, which could reduce the disease’s symptoms by producing a substance called N-acetylglucosamine from natural chitin resources. These bacteria could be transplanted into the human intestines, by the means of yoghurts or capsules. With this method it would be much easier to treat patients with Crohn’s disease in the future.
