Tell us about your team, your school!
Project
With the advent of synthetic biology and commercial usage of bacterial cells, one major concern is that rogue cells may escape their controlled environment, and alter the outside ecosystem. As humans, we have brought genetically modified organisms into systems for the purpose of fixing a problem, however things don’t always go as planned. The importance of a kill switch is for the control of the particular bacterial cells in question. The goal is to produce a way to terminate the bacteria, and therefore its function, to prevent further spread of the cells outside of the system.
The goal of this project is to produce a device that will cause the bacteria to self-destruct upon exposure to a factor in the environment not present in a laboratory setting. Our team will design and build a kill switch under the control of an inducible promoter. The self-destruction method will be determined later - we are evaluating several options such as lysis induction, production of an antibiotic or antimicrobial peptides, etc. After this device has been produced, the team will characterize its success by placing the bacteria in a situation which will simulate environmental release. The creation of a kill switch is imperative for acceptance of the idea of commercial usage of genetically modified organisms.
